To help the runaway slave Samuel, Zagor goes against the mercenaries of the Isthmian Guard, vigilantes under the orders of the governor and led by Ross Mc Gunn, known as "El diablo yanqui". Mc Gunn is allied with the sinister Mendoza and is determined to recapture the black man who escaped from his plantations. Samuel is headed towards a community of ex-slaves, led by a mysterious white man who calls himself Captain Mission, hidden in the jungle of Panama. Zagor, who must also cross the isthmus, decides to accompany him. With them is the merchant of Scottish origin Cullen Scott, who guides them into the tangle of the forest, where the Guaymì Indians also nest. But Captain Mission is also of great interest to McGunn: the Yankee is in fact convinced that he has found the mythical "treasure of Lima" which has been rumored in the region for years... and Samuel could take him to his secret hideout!

Along the road from the port of Callao to Lima, Zagor and Chico are attacked by a group of criminals intent on killing the lawyer Aguilar, who is traveling with them. Seeking refuge after the ambush, the trail of the Spirit with the Hatchet crosses with that of Barranco, the unscrupulous adventurer determined to reach a sacred city of the Incas hidden on the Andean peaks. To find her, the man does not hesitate to kidnap and take with him the beautiful Soledad, daughter of the curandera mama Jacinta. Meanwhile, Dexter Green and Yambo, having discovered important information in an ancient sixteenth-century codex, set off for Cuzco. And that's where Zagor is also headed, following in Barranco's footsteps...
